چکیده

An efficient embryogenic response (both direct and indirect) of two different varieties of sugarcane (CP 77,400 and BL-4) was obtained in this study. The results demonstrates that fresh inner whorl of leaves and shoot apical meristem of sugarcane were highly amenable to in vitro somatic embryogenesis (both direct and indirect). A combination of 3.0 mg/l 2,4-D with 0.25 mg/l BAP was more potent for direct embryo induction and growth in both the varieties of sugarcane. For indirect somatic embryogenesis the combination of 1.0 mg/l 2,4-D with 0.25 mg/l BAP showed best results in CP 77,400 while in BL-4 it was 1.0 mg/l 2,4-D with 0.5 mg/l BAP. The optimum temperature for callus induction and proliferation was found to be 27 ± 1°C. In CP 77,400, best shoot regeneration response was obtained on MS medium supplemented with 1.0 mg/l BAP. while in BL-4, MS medium containing 1.0 mg/l BAP + 0.5 mg/l Kinetin showed best organogenic

Effect of Salicylic Acid on Somatic Embryogenesis and Chlorogenic Acid Levels of Carrot (Daucus carota cv. Nantes) Explants

Many factors may influence the efficiency of somatic embryogenesis. This capability may be differ with regard to media compositions, genotype, tissue, organ ontology and the stage of differentiation. The effects of five salicylic acid concentrations (0, 25, 50, 75 and 100 mM) on different stages of carrot somatic embryogenesis were studied using petiole and root secondary phloem explants as sta...

SOMATIC EMBRYOGENESIS AND PLANTLET FORMATION IN DIFFERENT VARIETIES OF SUGARCANE (Sacchrum officinarum L.) HSF-243 AND HSF-245

An efficient protocol was developed for the induction of callus, somatic embryogenesis and regeneration response of two sugarcane varieties: HSF-243 and HSF-245, in this study. Callus was induced on young leaf explants cultured on Murashige and Skoog’s medium supplemented with different concentration of 2, 4-D singly or in combination with BAP. The highest percentage of callus induction was obs...

Induction of somatic embryogenesis in absence of exogenous auxin in cucumber (Cucumis sativus L.)

Induction of somatic embryogenesis in absence of exogenous auxin can clarify some basic concepts of somatic embryogenesis.  Explants from leaf and petiole of Cucumber (Cucumis sativus) were used in this experiment for somatic embryogenesis using three different modified B5 basal media; boron and 2,4-D but no IAA as control (I), boron and IAA but no 2,4-D (II) and no boron, 2,4-D and IAA (III). ...

Induction of Callus and Somatic Embryogenesis from Cotyledon Explants of Fagonia indica Burm.

To explore the potential for in vitro rapid regeneration of Fagonia indica Burm., cotyledon explants were obtained from 10-day old aseptically germinated seedlings that cultured on MS medium which supplemented with 1.0-5.0 µM 2,4-D. Callus was initiated from cotyledon explant on MS medium supplemented in all concentrations of 2,4-D investigated. 2,4-D was more effective than IAA and NAA for cal...

بررسی تأثیر برخی عوامل مؤثر بر جنین‌زایی و بلوغ جنین‌های رویشی در سویا (Glycine max)

Somatic embryogenesis is affected by several factors. In this research project, we studied the effect of explant size, wounding and desiccation treatments on somatic embryogenesis and their conversion into plantlet among three genotypes of soybean. The explants were sampled from immature embryos of soybean in three different sizes (3, 5 & 7 mm) with wounding treatment on half of each, and then ...
